Generalized q‐sampling imaging‐based fiber tracking measures of white matter tracts in cognitive impairment

چکیده
Background Diffusion magnetic resonance imaging (dMRI) is a non-invasive technique for exploring white matter fiber organization of the brain. As traditional diffusion tensor (DTI) methods may provide inaccurate descriptions local crossing and diverging fibers, we examined generalized q-sampling (GQI), novel method providing better directional quantitative information using dMRI data from community-dwelling older adults. Method We baseline 227 adults aged 71±8 on average Wake Forest Alzheimer’s Disease Research Center’s Clinical Core, who had year-1 follow-up global Dementia Rating (CDR) score 0 (indicates normal cognitive function) 0.5 (akin to MCI). The primary outcome measure was migration status determined by CDR increase/decrease between (mean difference = 13.9 months), shown in Figure. performed atlas-based tracking analysis GQ DSI Studio generate normalized anisotropy (NQA), DTI fractional (DTI_FA) restricted (RDI) measures several AD-related (WM) tracts: Corpus Callosum (CC), Fornix, Cingulum bundle/Parahippocampal tracts (see Table 2: CFP; CP; CPP), Uncinate Fasciculus (UF), Superior Longitudinal (SLF) Inferior Fronto-Occipital (IFOF). multinomial logistic regression (MLR) models adjusted age, sex, education, as standardized GQI-based indices sNQA, sRDI, sDTI_FA predictors. Result 1 lists demographics biomarker characteristics groups. MLR results 2 show significant associations 4 (Reversion) reference (stability cognition). Future work will validate these markers other groups (negative MCI stability).

Automated Detection of White Matter Fiber Tracts
White matter (WM) fiber bundles of the brain can be delineated by diffusion tractography utilizing anatomical regions-of-interest (ROI). These ROIs can specify seed regions in which tract generation algorithms are initiated. Interactive identification of such anatomical ROIs enables the detection of the major WM fiber tracts, but suffers from inter-rater and intra-rater variability, and is time...
متن کاملDamage to white matter fiber tracts in acute spatial neglect.
Previous statistical voxelwise lesion-behavior mapping (VLBM) studies have demonstrated that spatial neglect is associated with cortical and subcortical gray matter damage. However, it has also been suggested that the disorder may result from white matter injury. Our aim was to investigate the white matter connectivity in a large sample of 140 stroke patients. We combined a VLBM approach with t...
متن کاملA Unified Parametric Model of White Matter Fiber Tracts
We present a novel unified framework for explicitly parameterizing white fiber tracts. The coordinates of tracts are parameterized using a Fourier series expansion. For an arbitrary tract, a 19 degree cosine expansion is found to be sufficient to reconstruct the tract with an error of about 0.26 mm. By adding specific periodic constraints to open tracts, we can avoid using the sine basis. Then ...
متن کاملProbabilistic Clustering and Quantitative Analysis of White Matter Fiber Tracts
A novel framework for joint clustering and point-by-point mapping of white matter fiber pathways is presented. Accurate clustering of the trajectories into fiber bundles requires point correspondence determined along the fiber pathways. This knowledge is also crucial for any tract-oriented quantitative analysis. We employ an expectation-maximization (EM) algorithm to cluster the trajectories in...
متن کاملA test-retest reliability analysis of diffusion measures of white matter tracts relevant for cognitive control.
Recent efforts to replicate structural brain-behavior correlations have called into question the replicability of structural brain measures used in cognitive neuroscience. Here, we report an evaluation of test-retest reliability of diffusion tensor imaging (DTI) measures, including fractional anisotropy, mean diffusivity, axial diffusivity, and radial diffusivity, in several white matter tracts...
